I think you have some existing string and you want to check if that string exists in any row of the datatable by removing the space and convert the same to upper or lower case .Please check the below code for the same //Text needs to check var findText = testName ; //Linq query to convert the datatble row's value to upper case //and check with existing string var query = (from myRow in dt.AsEnumerable() where myRow.Field string >( Name ).Replace(    string .Empty).ToUpper() == findText.ToUpper() select myRow.Field string >( Name )).FirstOrDefault();

For Example: IEnumerable ds= from dt in TempTable .AsEnumerable() select dt.Field ( Temptable Columnname ); ds varaiable get list of string from temptable.ds value remove the space and convert the lowercase and check with text box value.
