I want bind data from sql db into combo box in vb??

First you are saying  select Student_branch...  in select statement and later you are saying .Add(dr(rollno)) . How does that suppose to work. Another general advice is try to stay away from DataAdapter DataSet and DataTable objects as much as possible. Instead use SqlDataReader which is read only by definition and you are not writing to data here. In addition you are saying cmbBranch.Items.Add and few lines below that you are also saying cmbBranch.DataSource. You are suppose to work with either of these not both. Remove the code adding to .Items collection. See if cmbBranch has DataBind() method if so call it then only data gets bound. Please post progress errors etc. in as much detail as possible.

In addition to answer given by Nitin Sontakke why don't you go with EF? That will organize your project structure far better than ADO.NET.
