Hi If you are using third party control or some custom control witch required registration in web.config then you have to add web.config file. For example you are using Ajax Control toolkit then it should be register in web.config or machine.config file.

yes the application will work without web.config but you will not be able to debug and the app will fail if you are using any information from web.config settings.
